Substantive content: 1.The frequencies of genotypes and alleles. The calculation of the allele frequencies in cases of complete dominance. Eugenics in the light of population genetics. The heterozygosities and other indices of genetic variability 2.Hardy – Weinberg genetic equilibrium. The use of HW law for genetic profiling. Forensic genetics for protection of the endangered species. The CITES convention 3.The effects of low numbers in populations. Genetic drift, inbreeding and inbreeding depression, mutational meltdown. The calculation of inbreeding coefficient from pedigrees for individuals and for populations 4.F statistics – the differentiation of populations, the gene flow and inbreeding 5.Protection plans, solving taxonomical problems. The species concept and practical approach to the problem. The barcoding idea. 6.Phylogeography and the units for protection within the species. The idea of ESU evolutionary significant unit and MU management unit 7.The minimum size of the viable population. IUCN categories and criteria for endangered species. Schedule for summer term 2024 1. Lecture – Hardy-Weinberg law, the deviations from HWE and their causes. Description of the genetic variability – basic parameters Lab – the calculations of the allele and genotype frequencies, HW equilibrium, parameters of genetic variability within population, gametic disequilibrium (LD) 22.02 2. Lecture – the methods for genetic variability research, protein electrophoresis and several methods for DNA research as RFLP, microsatellites, SNPs and DNA sequencing Lab – The red deer and sika deer – the example of hybridization research 29.02 3. Lecture – evolutionary forces, selection, migration, mutation and genetic drift. Mating systems and inbreeding Lab – calculation of the inbreeding coefficient from the pedigree. Inbreeding depression 7.03 4. Lecture – gene flow and fragmentation of the environment. F statistics Lab – calculation and interpretation of F statistics parameters 14.03 5. Lecture – conservation genetics – why genetics is important in extinction of species. Forensic genetics and DNA profiles Lab – the activity “who poached the moose?” showing the use of genetic markers for wildlife forensic genetics. The calculations for genetic profiles 21.03 6.
